Objective To establish a method for the determination of chlorogenic acid and magnelin in CangXin Nasal Drops. Methods For chlorogenic acid, HPLC method was used in the quantitative analysis using a Shim-Pack CLC ODS C18 (250×4.6 mm,5 μm) column. The mobile phase was acetonitrile-0.4% phosphoric acid (10:90) and detecting wavelength was 327nm, the flow rate was 1.0 ml/min. For magnelin, HPLC method was used in the quantitative analysis using a Hypersil C8 (250×4.6 mm,5 μm) column.. The mobile phase was acetonitrile-tetrahydrofuran-water(35:1:64) and detecting wavelength was 278nm, the flow rate was 1.0 mL/min. Results The calibration curve was linear in the range of 12.99~129.90 μg/mL(r=0.999 9) for chlorogenic acid and 19.67~196.70 μg/mL(r=0.999 9) for magnelin. The average recovery of chlorogenic acid was 99.28% and RSD was 1.15%(n=9), while, magnelin was 99.42% and RSD was 0.68%(n=9). The content of chlorogenic acid was 0.818 4 mg/mL, 0.818 5 mg/mL,0.818 9 mg/mL, while that of magnelin was 0.163 7 mg/mL, 0.163 4 mg/mL, 0.164 1mg/mL. Conclusion This method is simple, sensitive, accurate and available for the quality control of CangXin Nasal Drops. |
