目的 检测支气管哮喘患儿外周血单个核细胞中长链非编码RNA X染色体失活特异性转录本(lncRNA XIST)、微小RNA-124(miR-124)表达水平,并探讨其临床意义。方法 选取2020年1月至2021年3月因支气管哮喘于河南大学第一附属医院门诊及病房接受治疗168例患儿,其中急性发作期82例(急性发作组)、缓解期86例(缓解组);同期选取84例健康体检儿童作为对照组。实时荧光定量PCR法检测外周血单个核细胞中lncRNA XIST、miR-124水平,肺功能仪检测肺功能指标第一秒用力呼气量(FEV1),酶联免疫吸附法检测炎症因子白细胞介素(IL)-17、IL-1β、IL-10、肿瘤坏死因子-α(TNF-α)水平;Pearson法分析支气管哮喘患儿lncRNA XIST、miR-124与肺功能指标、炎症因子水平相关性。结果 与对照组比较,缓解组、急性发作组lncRNA XIST及IL-17、IL-1β、TNF-α水平升高,miR-124、FEV1、IL-10水平降低,差异有统计学意义(P<0.05);与缓解组比较,急性发作组lncRNA XIST及IL-17、IL-1β、TNF-α水平升高,miR-124、FEV1、IL-10水平降低,差异有统计学意义(P<0.05)。支气管哮喘患儿外周血单个核细胞lncRNA XIST与miR-124水平呈负相关(r=-0.668,P<0.001)。支气管哮喘患儿lncRNA XIST与IL-17、IL-1β、TNF-α呈正相关(P<0.05),与FEV1、IL-10呈负相关(P<0.05);miR-124与FEV1、IL-10呈正相关(P<0.05),与IL-17、IL-1β、TNF-α呈负相关(P<0.05)。结论 支气管哮喘患儿外周血单个核细胞lncRNA XIST高表达,miR-124低表达,二者可能参与支气管哮喘疾病的发生发展。 |

Objective To detect the expression levels of long non-coding RNA X inactive specific transcript (lncRNA XIST) and microRNA-124 (miR-124) in peripheral blood mononuclear cells of children with bronchial asthma, and to explore thesignificanceof its clinical features. Methods From January 2020 to March 2021, 168 children treated in the outpatient and ward of the First Affiliated Hospital of Henan University for bronchial asthma were selected. Among them, 82 cases were in acute attack (acute attack group) and 86 in remission (remission group); during the same period, 84 healthy children were selected as the control group. Real-time fluorescent quantitative PCR (qRT-PCR) was used to detect the levels of lncRNA XIST and miR-124 in peripheral blood mononuclear cells, a pulmonary function meter was used to detect the lung function index of forced expiratory volume in the first second (FEV1), and enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of inflammatory factors interleukin (IL)-17, IL-1β, IL-10, and tumor necrosis factor-α (TNF-α); Pearson method was used to analyze the correlation between lncRNA XIST, miR-124 and lung function indexes and inflammatory factor levels in children with bronchial asthma. Results Compared with the control group, the levels of lncRNA XIST, IL-17, IL-1β, and TNF-α increased in the remission group and the acute attack group, while the levels of miR-124, FEV1, and IL-10 decreased (P<0.05); compared with the remission group, the levels of lncRNA XIST, IL-17, IL-1β, and TNF-α in the acute attack group increased, while the levels of miR-124, FEV1, and IL-10 decreased (P<0.05). Peripheral blood mononuclear cell lncRNA XIST in children with bronchial asthma was negatively correlated with the level of miR-124 (r=-0.668, P<0.001). In children with bronchial asthma, lncRNA XIST was positively correlated with IL-17, IL-1β, and TNF-α (P<0.05), and negatively correlated with FEV1 and IL-10 (P<0.05); miR-124 was positively correlated with FEV1 and IL-10 (P<0.05), and negatively correlated with IL-17, IL-1β, and TNF-α (P<0.05). Conclusions In children with bronchial asthma, lncRNA XIST is highly expressed in peripheral blood mononuclear cells, while miR-124 is lowly expressed. The two may be involved in the occurrence and development of bronchial asthma. |
