      免疫疗法是治疗肺腺癌 (LUAD)的有效手段,但缺乏预测反应及耐药分析是其面临的巨大挑战。本文通过阐述铜死亡发生机制,围绕铜死亡相关基因与LUAD预后、LUAD免疫治疗评估两个方面进行论述,展开对LUAD患者总生存期预测、免疫治疗反应、化疗治疗反应、风险评估、干性特征等因素的探讨,总结铜死亡预测肺腺癌预后规律及指导LUAD免疫治疗的作用,揭示铜死亡相关基因在LUAD中突变频率不确切等问题,为今后铜死亡关于LUAD的治疗和预后研究提供新方向。
英文摘要:
      Unlike apoptosis, necrotic apoptosis, and iron death in recent years, cuproptosis is a novel form of cell death dependent on copper. Its main mechanism is the direct binding of copper to the fatty component of the TCA cycle in human cells, resulting in the accumulation of fatty proteins and the subsequent loss of iron sulfur cluster (ISC) proteins. Eventually, the protein becomes toxic and causes cell death. Lung cancer is the deadliest malignant tumor in the world. Lung adenocarcinoma (LUAD), as the most common tissue type of lung cancer, is highly invasive and lethal, with an overall survival of less than 5 years. In recent years, the rapid rise of immunotherapy-immune checkpoint blocking (ICB) therapy has revolutionized human cancer treatment, including LUAD patients. ICB is a recognized effective strategy for treating LUAD patients, but the lack of effective biomarkers to predict response or resistance is a major challenge for ICB therapy. Therefore, it is urgent to explore biomarkers with effective predictive significance to guide immunotherapy. Recent studies have found that there is a close relationship between cuproptosis and LUAD. This paper discusses cuproptosis related genes, LUAD prognosis and LUAD immunotherapy evaluation by describing the mechanism of cuproptosis. It is hoped to provide reference for improving the mechanism of cuproptosis in LUAD and guiding the clinical treatment of LUAD.
