目的 研究核因子(NF)κB介导的PTX3和CRP通路,是否可作为治疗妊高症的潜在靶点并分析其临床意义。方法 选取2012年3月至2014年12月收治的妊高症患者(NH BPEP标准)60例作为观察组,其中妊高症20例[血压(131.2±17.4)/(79.7±10.0)mmHg],轻度先兆子痫20例[血压(141.2±15.6)/(89.5±11.2)mmHg],重度先兆子痫20例[血压(150.3±17.6)/(97.6±14.8)mmHg];选取同期入院血压正常的晚期妊娠妇女20例作为对照组,采用全自动生化分析仪检测两组对象外周单个核细胞PTX3和CRP水平,采用免疫印迹杂交检测NF-κB的水平;加入NF-κB抑制剂(PTDC),观察PTX3和CRP表达水平的变化;同时观察其与患者血压波动的关系。结果 观察组PTX3和CRP随血压升高而升高,均明显高于对照组,组内、组间差异均有统计学意义(P<0.05);加入PTDC后,PTX3和CRP水平均降低,与对照组比较,差异无统计学意义(P>0.05)。重度先兆子痫患者NF-κB表达最高[(25.8±4.9)%],应用PTDC后降低[(13.9±4.9)%],与对照组[(13.5±3.6)%]比较,差异无统计学意义(P>0.05)。结论 血浆PTX3和CRP是核因子κB的下游因子,在妊高症患者中表达升高,该通路可作为药物治疗妊高症的潜在靶点。 |

Objective To investigate the nuclear factor κB mediated plasma five poly 3(PTX3)-reactive protein(CRP) pathways in patients with hypertension in pregnancy. Methods Sixty cases of patients with hypertensive disorder complicating pregnancy who were admitted in our hospital from March 2012 to December 2014 were selected as the observation group, in which 20 cases had pregnancy induced hypertension,[BP (131.2±17.4)/(79.7±10.0)mmHg], 20 cases had mild preeclampsia[BP(141.2±15.6)/(89.5±11.2)mmHg], and 20 cases had severe preeclampsia[BP(150.3±17.6)/(97.6±14.8)mmHg]. Another 20 cases of normal late pregnant women with the same admission were selected as the control group, then the automatic biochemical analyzer was used to detect the peripheral mononuclear cells of plasma five poly3 and CRP level, and Western blot was used in detection of the level of NF-κB. Then NF-κB inhibitor was added, PTX3 and CRP level was observed, and at the same time its relation with blood pressure fluctuation was observed. Results PTX3 and CRP in the observation group were increased with the rise of blood pressure, which were significantly higher than those of the control, and the difference within groups and between groups had statistically significant difference. After addition of PTDC, the two indexes were reduced, down to the level of control group, and compared with II degrees and Ⅲ degrees, the difference was statistically significant. NF-κB level increased with elevated blood pressure. Conclusion Plasma five poly 3 and C reactive protein is a downstream factor nuclear factor kB expression, elevated in patients with hypertension of pregnancy, and the channel may be used as a potential target for the treatment of pregnancy induced hypertension with drugs. |
