目的 建立兔开放性脊髓损伤模型,观察海水浸泡对兔开放性脊髓损伤后TNF-α、VEGF表达的影响。方法 采用改良Allenś打击器造兔脊髓损伤模型。选用日本大耳兔,体质量2.5~3 kg,雌雄不拘。根据随机数字表,将60只大耳白兔随机分为两组,实验组:海水浸泡组(切除椎板并造脊髓损伤模型,海水浸泡60 min,n=30),对照组:单纯损伤组(切除椎板并造脊髓损伤模型,不浸泡,n=30)。分别于致伤后1、6、12、24、48小时5个时间点取伤段脊髓,光镜下观察伤段脊髓组织病理形态,免疫组织化学染色和计算机图形分析技术半定量化检测TNF-α、VEGF表达。结果 两组脊髓组织均发生了创伤性水肿,但水肿高峰期的时间点不同,单纯损伤组致伤后6小时达水肿高峰,而海水浸泡组于致伤后12小时达水肿高峰。免疫组化检测结果显示TNF-α、VEGF在两组中均有阳性表达,但两组在各时间点表达强度有差异(P < 0.05),且海水浸泡组TNF-α、VEGF表达的高峰与对照组不同。结论 海水浸泡早期延迟了创伤性脊髓损伤的病理改变,但最终发生的脊髓损害较对照组加重。TNF-α、VEGF的高表达与损伤有着密切关系,可作为海水浸泡脊髓损伤预后判断、伤情评估的指标。 |

Objective To establish model of spinal cord injury with seawater immersion,observe the influence on neural cell expres-sion of TNF-α and VEGF. Methods The rabbit model of spinal cord injury were built based on modified Allenś method. According to random digital table,60 rabbits were randomly divided into two groups, experimental group:cut vertebral plate and damaged spinal cord with seawater immersion(n=30,and the immersion time is 60 minutes). Control group:cut off vertebral plate and damaged spinal cord without seawater immersion(n=30). Two groups randomly divided into five groups based on 1 hour,6 hours,12 hours,24 hours,48 hours after im-mersion. The animals were sacrificed at every timepoint,and pathological of spinal cord was studied by light microscopy, immunohistochemis-try was used to test the expression of TNF-α and VEGF. Result The traumatic spinal cord edemous peak of control group at the timepoint of 6 hours,but the peak of experimental group was postponded to the 12 hours. Immunohistochemistry showed positive expression of TNF-α, VEGF are in two groups,but the expression strength of two groups has notable diffence in each timepoint(P < 0.05). And the expression peak of TNF-α, VEGF is diffrent in two groups. Conclusion Seawater immersion delayed traumatic spinal cord injury in early stage, but it aggravated spinal cord injury finally. The high expression of TNF-α, VEGF indicated a close relationship with injury, these two kind of pro-teins can be used as judgement of prognosis and injury assessment of spinal cord injury with seawater immersion. |
