      目的 研究白藜芦醇(RSV)在大鼠肾脏缺血再灌注损伤(IRI)模型中降低炎症反应及抗凋亡作用。方法 将18只SD大鼠随机分成假手术组(Sham组)、IRI组和IRI+RSV组),每组6只,后两组建立IRI组模型,治疗组于术前2天、术前1天和术后第1天给予RSV(0.23μg/kg)灌胃。各组大鼠分别于术后36小时、48小时、第5天采集血标本及肾脏标本,评价肾功能和肾组织病理学损伤,采用原位缺口末端标记法(TUNEL)检测肾小管上皮细胞凋亡情况,采用实时定量反转录聚合酶链式反应(RT-qPCR)检测肾脏组织相关炎性细胞因子水平。结果 在肾脏IRI早期,与IRI组相比,IRI+RSV组大鼠血肌酐和尿素氮的水平明显降低,病理损伤较轻,TUNEL显示肾小管细胞凋亡(阳性细胞)减少,相关炎性细胞因子的表达水平也明显降低。结论 RSV可以通过降低大鼠IRI过程中的炎性反应,并起到抗凋亡的作用,进而发挥对肾脏的保护作用。

      Objective To investigate the protective effects of resveratrol on kidneys exposed to ischemia-reperfusion(IR) injury in early stage in a ratmodel by anti-inflammation and anti-apoptosis.Methods Eighteen SD rats were randomly divided into sham group, IR group and IR+RSV-treated group with six rats in each group. IR model was established in the last two groups by intragastric administration of resveratrol(RSV) (0.23 μg/kg) preoperative 2 d, preoperative 1 d and postoperative 1 d.Blood and renal samples in three groups were collected postoperative 36 h, 48 h and 5 th day for the assessment of renal function and pathological injury of kidney. Renal cell apoptosis was detected by TdT-mediated dUTP nick end labeling (TUNEL) assay. The expression of some related cytokines was detected by RT-qPCR.Results After RVS treatment, the serum creatinine and urea nitrogen level decreasedwith lighter pathological damage compared with IR group. Apoptotic renal cells in IR+RSV group also declined compared with IR group.Conclusion RSV protects renal function probablyby inhibiting cell apoptosis and reducing inflammation.
