      目的 探讨18F-FMISO、18F-FDG micro PET/CT显像评价裸鼠MDA-MB231乳腺癌移植瘤中肿瘤乏氧与增殖状态的可行性。方法 16只裸鼠MDA-MB231乳腺癌移植瘤模型先后进行18F-FDG及18F-FMISO PET/CT显像并采集肿瘤最大标准化摄取值[SUVmax (FDG)、SUVmax (FMISO)],所有模型显像结束后立即处死,取肿瘤组织行HIF-1α及Ki67免疫组织化学染色,将SUVmax值与HIF-1α、Ki67的表达水平进行相关性分析。结果 18F-FMISO平均SUVmax值1.69,18F-FDG平均SUVmax值5.80。SUVmax (FMISO)与HIF-1α、Ki67的表达均呈正相关性(r=0.74、0.52,P<0.05)。SUVmax (FDG)与Ki67的表达呈正相关性(r=0.86,P<0.05),与HIF-1α表达无相关性。HIF-1α与Ki67蛋白的表达呈正相关性(r=0.60,P<0.05)。结论 18F-FMISO PET/CT显像可同时对活体肿瘤进行乏氧及增殖的可视化监测,18F-FDG可提供有效的肿瘤增殖显像,但不能作为有效的肿瘤乏氧显像剂。

      Objective To investigate the feasibility of18F-FMISO and 18F-FDG Micro PET/CT imaging in evaluating tumor hypoxic and cell proliferation inMDA-MB231 breast cancer in nude mice. Methods Sixteen nude mice bearing human breast cancer(MDA231 cell line) xenografttumors were monitored with 18F -FDG PET/CT and 18F -FMISO PET/CT imaging.The SUVmax(FMISO) and SUVmax(FDG) were collected respectively. Mice were randomly sacrificed after PET/CT imaging, then immunohistochemical staining was used to assess HIF-1α and Ki67 expression. We investigated the correlations of different PET tracers' uptake with HIF-1α and Ki67expression.Results The averageof SUVmax(FMISO)was 1.69, while the averageof SUVmax(FDG) was 5.80. Both the expression of HIF-1α and Ki67 wererelated with SUVmax(FMISO)(r=0.74, P<0.05, r=0.52, P<0.05, respectively). SUVmax(FDG)was significantlycorelated with Ki67(r=0.86, P<0.05). No correlation was detected between SUVmax(FDG)and HIF-1αexpression.The expression of HIF-1α was also correlated with Ki67(r=0.60, P<0.05)Conclusion The uptake of FMISOisclosely correlated with the expressions of Ki67 and HIF-1α,18F-FMISO PET/CT plays an important role in visual detection of tumor hypoxic and cell proliferation. The uptake of FDG issignificantlycorrelated with Ki67, which can reflect tumor proliferation effectively, but not as an effectively hypoxic imaging agent.
