      目的 探究刺猬因子(Hedgehog)信号通路分子超音速刺猬蛋白(Shh)及其下游转录因子叉头框转录因子M1(FoxM1)和神经胶质瘤相关癌基因同源物1(Gli1)在宫颈癌中的表达水平及其与临床病理特征的关系。方法 选取遂宁市中心医院2016年1月至2016年8月收治的50例宫颈癌患者宫颈黏膜组织作为观察组,选取30例同期收入的宫颈黏膜组织正常的阴道炎患者作为对照组,应用免疫组化检测两组对象宫颈黏膜组织中Shh、FoxM1、Gli1的表达情况,分析宫颈癌组织中Shh、FoxM1、Gli1蛋白表达与临床病理特征之间的关系。结果 Shh、FoxM1、Gli1蛋白在宫颈癌组织中的表达阳性率分别为86%、90%、92%,较正常宫颈组织升高,差异有统计学意义(P<0.05);Shh、FoxM1、Gli1蛋白与患者年龄、肿瘤大小、浸润深度均无显著相关性(P>0.05);Spearman相关性分析显示,Shh在宫颈癌中的表达与组织分化程度、FIGO分期、淋巴结转移相关(P<0.05);FoxM1只与淋巴结转移相关(P<0.05);Gli1的表达水平与宫颈癌组织的分化程度、淋巴结转移相关(P<0.05);Shh、FoxM1、Gli1三者之间互为正相关(P<0.05)。结论 Shh、FoxM1、Gli1在宫颈癌组织中呈高表达状态,Shh和Gli1蛋白与宫颈癌组织分化程度有关,而FoxM1蛋白只与宫颈癌是否发生淋巴结转移有关,此三者的检测,有助于宫颈癌的早期诊断,可作为预测宫颈癌发生发展的指标。

      Objective To explore the expressionlevelsofsonic hedgehog (Shh) proteinin the Hedgehog signaling pathwayand its downstream transcription factors forkhead box transcription factor M1 (FoxM1) and glioma associated oncogene homolog 1 (Gli1)in those patients with cervical cancer, and theirrelationswith the clinicopathological features. Methods 50 cases of cervical cancer patients with abnormalcervical mucosa, ever treated in our hospital from Jan 2016 to Aug 2016,were enrolledas the research group,while 30 cases of vaginitiswomen with normal cervical tissues were selected simultaneouslyas the control group. Theexpression levels of Shh, FoxM1 and Gli1 proteinsin the cervical mucosa samples taken from all patients were detected by immunohistochemical staining, and the relationship between levels of Shh, FoxM1 and Gli1proteins in the research groupand their clinicopathological features of cervical cancer were analyzed. Results The positive expression rates of Shh, FoxM1 and Gli1 in the cervical cancer tissues were 86%, 90%and 92%,respectively, which wereallsignificantly higher than thoseinthe normal cervical tissues (P<0.05).The Shh, FoxM1 and Gli1 proteinshad no significant association with age, tumor size, depth of invasion of these patients(P>0.05). Spearman correlation analysis showed that the Shh expressionwas associated with degree of differentiation, FIGO stages and lymph node metastasis in the cervical cancer (P<0.05),but FoxM1 was only associated with lymph node metastasis (P<0.05), and Gli1 was associated with degree of differentiationand lymph node metastasis (P<0.05).The correlation analysis furtherindicated thatthe expression of Shh, FoxM1and Gli1 proteins weremutually positively correlated (allP<0.05). Conclusion The expression levels of Shh, FoxM1 and Gli1 proteinsin the cervical cancer tissues are higher, and the expression of Shh and Gli1are associated with degree of differentiation of cancer tissues, but the FoxM1 expression is only correlated tolymph node metastasis. Detection of the three indicatorswould be helpful to early diagnose the cervical cancer, and to predict its occurrence and development.
