目的 研究豚鼠近视模型中屈光度眼轴长度变化及后极部巩膜中Ⅰ型胶原纤维的变化。方法 取出生1周的豚鼠75只,利用6号半透明乳胶气球作为头套诱导形觉剥夺性近视(FDM)动物模型。随机分为空白对照组、形觉剥夺(FDM)组和自身对照组。分别测量记录实验豚鼠不同时空点的双眼屈光度及眼轴长度。形觉剥夺实验组分为遮盖2周、遮盖4周、遮盖4周去遮盖1周及遮盖6周4个亚组,左眼为遮盖眼(FDM组),采用半透明面罩遮盖诱导形觉剥夺性近视,右眼不予处理(自身对照组)。对遮盖前后实验组和对照组双眼屈光度、眼轴长度进行测量,并对后极部巩膜中Ⅰ型胶原进行免疫组化分析。结果 FDM组由遮盖前远视(+2.09±0.31)D逐渐变成遮盖6周时近视(-7.07±0.56)D,眼轴也由遮盖前(5.93±0.39)mm到6周时(7.99±0.32)mm,且后极部巩膜中Ⅰ型胶原含量也逐渐降低。结论 形觉剥夺法可成功制造豚鼠实验性近视模型,在豚鼠形觉剥夺性近视的形成过程中伴随着眼球近视度数逐渐增加和眼轴长度不断增长,且后极部巩膜中Ⅰ型胶原随着近视度数加深逐渐降低。 |

Objective To study the changes of diopter and axial length in guinea pig myopia model, and the changes of type I collagen fibers in the posterior sclera. Methods Seventy-five guinea pigs, one week old, were used to induce a form-deprivation myopia (FDM) animal model using a translucent latex balloon No. 6 as a hood. The animals were randomizedinto blank control group, form deprivation (FDM) group and self-control group. The diopter and axial length of binocular were measured at different time points in guinea pigs. The morphological deprivation experiment was divided into four groups:shaded for 2 weeks, shaded for 4 weeks, shaded for 1 week and shaded for 6 weeks; the left eye was shaded (FDM group), myopiawas inducedand the right eye wasnot treated (self-control group). The diopter and axial length of both eyes in experimental group and control group before and after the masking were measured, and type Ⅰ collagen in the sclera of posterior pole was immunohistochemically analyzed. Results The guinea pigs of FDM group gradually changed from hyperopia (+2.09±0.31) D to myopia (-7.07±0.56) D, the axial length was also changed from (5.93±0.39) mm to (7.99±0.32) mm, and the posterior pole sclera type Ⅰ collagen content was also gradually reduced. Conclusion Guinea pig forms the deprivation myopia process accompanied by the changes of refraction and axial length, and posterior pole sclera type Ⅰ collagen gradually decreaseswith myopia deepening. |
