目的 探讨人过氧化物氧化还原酶-1(PRDX1)在胃癌中的表达情况及其与胃癌临床病理特征和预后之间的关系。方法 采用实时荧光定量聚合酶链反应(qRT-PCR)和蛋白质印迹法(Western Blot)检测21例胃癌与相应癌旁组织中PRDX1 mRNA和蛋白的表达水平,并应用免疫组化法检测105例胃癌和癌旁组织中PRDX1蛋白的表达水平,以分析其表达情况与临床病理特征及预后的关系。结果 qRT-PCR和Western Blot检测发现,PRDX1在胃癌组织中的mRNA和蛋白的表达水平均高于癌旁组织,差异有统计学意义(P<0.05)。免疫组织化学检测结果表明,PRDX1蛋白水平在胃癌组织中高于癌旁组织,差异有统计学意义(P<0.05)。不同肿瘤分化程度、浸润深度、淋巴结转移情况、TNM分期间PRDX1蛋白表达水平差异有统计学意义(P<0.05)。Kaplan-Meier生存分析显示,PRDX1蛋白的过表达与总生存时间及无病生存时间较短相关。COX多因素分析显示,PRDX1是胃癌独立的预后影响因子。结论 胃癌组织中PRDX1蛋白的表达明显升高,PRDX1蛋白高表达可能促进了胃癌的发生和发展,可能做为胃癌的早期诊断及预后评估的新型生物学指标。 |

Objective To investigate the expression of human peroxide oxidoreductase-1 (PRDX1) in gastric cancer and its relationship with the clinicopathological characteristics and prognosis of gastric cancer.Methods Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) and Western blot were used to detect the expression level of PRDX1 mRNA and protein in 21 cases of gastric cancer and adjacent tissues, and 105 cases of gastric cancer and gastric cancer were detected by immunohistochemistry. The expression level of PRDX1 in adjacent cancer tissues was used to analyze the relationship between its expression and clinicopathological characteristics and prognosis.Results The mRNA and protein expression levels of PRDX1 in gastric cancer tissues were higher than those in adjacent tissues, and the difference was statistically significant (P<0.05). Immunohistochemical results showed that PRDX1 levels were higher in gastric cancer tissues than in adjacent tissues, and the difference was statistically significant (P<0.05). Chi-square analysis showed that high expression of PRDX1 was significantly associated with tumor differentiation (P<0.05), depth of invasion (P<0.05), lymph node metastasis (P<0.05), and TNM stage (P<0.05). Kaplan-Meier survival analysis showed that overexpression of PRDX1 was associated with shorter overall survival and disease-free survival. Multivariate analysis showed that PRDX1 was an independent prognostic factor for gastric cancer.Conclusion PRDX1 expression in gastric cancer tissuessignificantly increases. The high expression of PRDX1 may promote the occurrence and development of gastric cancer,whichmay be used as a new biological indicator for the early diagnosis and prognosis evaluation of gastric cancer. |
