      目的 探究长链非编码RNA肺腺癌转移相关转录本1(LncRNA MALAT1)、微小RNA-206(miR-206)在子痫前期(PE)孕妇血浆外泌体中的表达水平及临床意义。方法 选取2017年1月至2019年12月在郑州市妇幼保健院住院分娩的PE孕妇42例为PE组,另选取同期在本院分娩的健康孕妇42例为对照组。根据入组PE患者病情严重程度分为轻度组22例,重度组20例。检测所有研究对象血浆外泌体LncRNA MALAT1、miR-206、血管内皮生长因子(VEGF)表达水平并进行组间比较。采用Pearson法分析PE孕妇血浆外泌体中LncRNA MALAT1、miR-206和VEGF的相关性,logistic回归模型分析PE发生的影响因素,采用受试者工作特征曲线(ROC)分析血浆外泌体LncRNA MALAT1、miR-206表达水平对PE发生的预测价值。结果 与对照组比较,PE组患者血浆外泌体miR-206水平明显升高,LncRNA MALAT、VEGF水平明显降低(P<0.05)。LncRNA MALAT1、VEGF水平随PE患者病情加重有降低趋势,miR-206水平有升高趋势(P<0.05)。高miR-206水平、低LncRNA MALAT1水平是PE发生的独立危险因素(P<0.05)。LncRNA MALAT1、miR-206联合检测预测PE发生的曲线下面积为0.872,敏感度为92.86%。结论 PE患者血浆外泌体中LncRNA MALAT1呈低表达、miR-206呈高表达,两者均是PE发生的危险因素,二者联合检测对PE的诊断价值较高。

      Objective To investigate the expression levels and clinical significance of long non-coding RNA metastasis-associated lung adenocarcinoma transcript 1 (LncRNAMALAT1) and microRNA-206 (miR-206) in plasma exosomes of preeclampsia (PE) pregnant women. Methods From January 2017 to December 2019, 42 PE pregnant women in Zhengzhou Maternal and Child Health Hospital were selected as the study objects (PE group), and another 42 healthy pregnant women who delivered in the same hospital at the same time were selected as the control group. According to the severity of PE patients, they were divided into mild group (22 cases) and severe group (20 cases). The expression levels of LncRNAMALAT1, miR-206 and vascular endothelial growth factor (VEGF) in plasma exosomes were detectedand compared between the two groups. Pearson method was used to analyze the correlation of LncRNA MALAT1, miR-206 and VEGF expression levels in plasma exosomes of PE pregnant women. Logistic regression model was used to analyze the influencing factors of PE. Receiver operating characteristic (ROC) curve was used to analyze the predictive value of LncRNA MALAT1, miR-206 expression levels in plasma exosomes on PE. Results Compared withcontrol group, the expression level of miR-206 in plasma exosomes in PE group significantly increased, and the expression levels of LncRNA MALAT1 in plasma exosomes significantly decreased (P<0.05).LncRNA MALAT1 level tended to decrease with the aggravation of PE patients, while MiR-206 level tended to increase (P<0.05).High miR-206 level and low LncRNAMALAT1 levels were independent risk factors for PE (P<0.05).The area under the curve of PE predicted by LncRNAMALAT1 and miR-206 was 0.872, and the sensitivity was 92.86%. Conclusions PEpatients have low expression of LncRNAMALAT1and high expression of miR-206 inplasmaexosomes,both of which areriskfactorsfor PE, and the combined detection of the two has a high diagnostic value for PE.
