目的 通过检测黏着斑激酶(FAK)和增殖细胞核抗原(PCNA)在口腔白斑病(OLK)中的表达水平,探讨FAK对细胞增殖的影响。方法 选择2017年1月至2019年12月在安徽医科大学第一附属医院进行OLK手术切除的41例患者作为试验组,根据病理诊断,将试验组41例OLK标本分为OLK上皮单纯增生组(n=10)、OLK上皮轻度异常增生组(n=11)、OLK上皮中度异常增生组(n=13)和OLK上皮重度异常增生组(n=7)。另同期选取10例进行囊肿切除术以及智齿拔除术患者的正常口腔黏膜组织作为对照组,采取免疫组织化法检测各组患者的FAK和PCNA表达;同时采用逆转录-聚合酶链反应法检测各组患者FAK mRNA表达。结果 5组FAK蛋白表达量的差异有统计学意义(P<0.05),对照组与OLK上皮单纯增生组相比,差异无统计学意义(P>0.05),其余各组两两相比,差异均有统计学意义(P<0.05);5组PI指数的差异有统计学意义(P<0.05),对照组与OLK上皮单纯增生组相比,差异无统计学意义(P>0.05),其余各组两两相比,差异均有统计学意义(P<0.05);5组FAK mRNA表达量的差异无统计学意义(P>0.05),FAK蛋白表达与PI呈线性正相关(r=0.964,P<0.05)。结论 FAK和PCNA在OLK上皮异常增生的患者中表达上调,二者的表达水平与OLK的癌变过程有关,可能作为评价OLK癌变过程的潜在标记物。 |

Objective To investigate the effect of adhesion plaque kinase(FAK) on cell proliferation via detection of the expression levels of FAK and proliferating nuclear antigen (PCNA) in oral leukoplakia (OLK). Methods A total of 41 patients who underwent OLK surgical resection in the First Affiliated Hospital of Anhui Medical University from January 2017 to December 2019 were selected as the experimental group. According to the pathological diagnosis, the 41 OLK specimens in experimental group were divided into simple hyperplasia group (n=10), mild dysplasia group (n=11), moderate dysplasia group (n=13) and severe dysplasia group (n=7).In addition, normal oral mucosal tissues from 10 patients undergoing cystectomy and wisdom tooth extraction in our hospital during the same period were selected as the control group. The expressions of FAK and PCNA in each group were detected by immunohistochemistry. The mRNA expression of FAK in each group was detected by reverse transcription-polymerase chain reaction. Results The difference in FAK protein expression of the five groups was statistically significant (P<0.05), and there was no statistically significant difference between control group and OLK epithelial hyperplasia group (P>0.05), and there was statistically significant difference between the other groups (P<0.05). There was statistically significant difference in PI index between the five groups (P<0.05), and there was no statistically significant difference between control group and OLK epithelial hyperplasia group (P>0.05), and there was statistically significant difference between the other groups (P<0.05). There was no significant difference in the mRNA expression of FAK among the five groups(P>0.05), and FAK protein and PI showed a positive correlation in oral leukoplakia(r=0.964, P<0.05). Conclusions The expressions of FAK and PCNA are up-regulated in patients with OLK epithelial dysplasia, and the expression levels of both are related to the canceration process of OLK, which may be used as potential markers to evaluate the canceration process of OLK. |
