      目的 探讨类RAS激活物2(RASAL2)在胃癌中的表达及其临床意义。方法 通过Oncomine、GEPIA和Kaplan-Meier Plotter数据库分析RASAL2在胃癌中的基因表达及其与预后的关系。通过cBioPortal数据库分析胃癌患者中RASAL2基因变异情况及其与预后的关系。体外培养胃癌细胞MGC-803,通过siRNA转染沉默RASAL2,通过CCK-8、细胞划痕实验及Transwell侵袭实验检测MGC-803细胞增殖、迁移与侵袭。结果 胃癌组织中RASAL2基因表达高于正常组织(P<0.05)。与低表达组相比,RASAL2高表达组患者总体生存率降低(P<0.05);亚组分析显示,RASAL2高表达与Ⅰ期(HR=6.22,P<0.05)、Ⅲ期(HR=1.6,P<0.05)及中分化(HR=4.74,P<0.05)胃癌患者不良预后相关。胃癌患者中RASAL2基因总体变异率为7%,且变异组无病生存率高于非变异组(P<0.05)。沉默RASAL2可抑制MGC-803细胞增殖、迁移及侵袭(P<0.05)。结论 RASAL2在胃癌组织中表达增高,高表达RASAL2与胃癌患者不良预后相关,沉默RASAL2可抑制胃癌MGC-803细胞增殖、迁移和侵袭能力。

      Objective To explore the expression of RAS protein activator like2(RASAL2) and its clinical significance in gastric cancer. Methods The gene expression of RASAL2 and its relationship with prognosis in gastric cancer were analyzed using the Oncomine,GEPIA and Kaplan-Meier Plotter databases. The mutations of RASAL2 gene and their relationship with prognosis were analyzed through the cBioPortaldatabase.Gastric cancer cells MGC-803 were cultured in vitro. RASAL2 was knocked down by siRNA transfection. The proliferation, migration and invasion of MGC-803 cells were detected by CCK-8,cell scratch-healing assay and Transwell invasion test, respectively. Results The gene expression of RASAL2 was up-regulated in gastric cancer tissues compared to that in normal tissues (P<0.05).The overall survival rate was lower in the patients with higher expression levels of RASAL2 (P<0.05);subgroup analyses were conducted, which showed that higher expression levels of RASAL2 were related to poor prognosis in patients with stage Ⅰ(HR=6.22,P<0.05),stage Ⅲ(HR=1.6,P<0.05) and moderately differentiated (HR=4.74,P<0.05) gastric cancer. The overall alteration rate of RASAL2 gene in gastric cancer was 7%, and the disease-free survival rate of the mutation group was higher than that of the non-mutation group(P<0.05).Knockdown of RASAL2 could inhibit the proliferation, migration and invasion of MGC-803 cells(P<0.05). Conclusions The expression of RASAL2 increases in gastric cancer tissues. High expression of RASAL2 is associated with poor prognosis in gastric cancer. Silencing RASAL2 can inhibit the proliferation, migration and invasion of gastric cancer MGC-803cells.
