Objective To investigate the bacteriostatic effect of platelet-rich plasma(PRP) and its composite silver ion in vitro, and to study the effect of silver ion on the release of growth factors by PRP. Methods PRP and silver ion solution were prepared, and PBS buffer was used as a negative control. The growth inhibition and the duration of antibacterial effect of PRP, silver ion and their composite on Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa were observed at 37℃. The concentrations of platelet-derived growth factor(PDGF),transforming growth factor-β(TGF-β), epidermal growth factor(EGF) and vascular endothelial growth factor(VEGF) of PRP alone and combined with silver ion were measured by ELISA. Results At 37℃, compared with the control group, the growth of Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa in the silver ion group and the PRP combined with silver ion group was inhibited in culture dishes.For Staphylococcus aureus and Escherichia coli, the antibacterial effect of the two materials could last for 36 hours, and the antibacterial effect on Pseudomonas aeruginosa began to decline after 24 hours. There was no significant difference in the concentration of PDGF, TGF-β, EGF and VEGF between PRP combined with silver ion group and PRP group(P=0.499, 0.362, 0.593, 0.794). Conclusions The combination of PRP and silver ions not only retains the antibacterial ability of silver ions, but also has no effect on the ability of PRP to release growth factors such as PDGF, TGF-β, EGF and VEGF. |
