| 目的 探讨组蛋白赖氨酸甲基转移酶2D(KMT2D)、膜联蛋白A7(ANXA7)、趋化素样因子超家族6(CMTM6)在结肠癌中的表达及其与患者根治术后复发转移的关系。方法 选取2018年6月至2021年6月邯郸市第一医院进行根治术的200例结肠癌患者,收集癌组织及癌旁组织,根据术后是否复发转移分为复发转移组(n=86)和未复发转移组(n=114)。通过免疫组化及实时荧光定量-聚合酶链式反应检测KMT2D、ANXA7、CMTM6蛋白表达情况及相对表达量,收集分析一般临床资料,Logistic回归分析术后复发转移的影响因素,受试者工作特征曲线分析KMT2D、ANXA7、CMTM6对术后复发转移的诊断价值。结果 KMT2D、ANXA7、CMTM6蛋白在结肠癌组织中的阳性表达率高于癌旁组织(P<0.05);复发转移组的TNM分期Ⅲ+Ⅳ期、低分化程度以及神经侵犯患者占比、KMT2D、ANXA7、CMTM6 mRNA相对表达量高于未复发转移组(P<0.05);TNM分期Ⅲ+Ⅳ、低分化程度、神经侵犯以及KMT2D、ANXA7、CMTM6 mRNA水平升高均是影响患者术后复发转移的危险因素(P<0.05);KMT2D、ANXA7、CMTM mRNA 3者联合预测根治术后复发转移的曲线下面积均高于KMT2D(Z=3.233,P=0.001)、ANXA7(Z=2.923,P=0.004)、CMTM6(Z=2.037,P=0.042)单独预测。结论 KMT2D、ANXA7、CMTM6 mRNA及蛋白在结肠癌中表达升高,与患者根治术后复发转移具有一定关系。 |

| Objective To investigate the expression of histone-lysine N-methyltransferase 2D(KMT2D), annexin A7(ANXA7), and chemokine-like factor superfamily 6(CMTM6) in colon cancer and their relationship with postoperative recurrence and metastasis in patients. Methods From June 2018 to June 2021, 200 colon cancer patients who underwent radical surgery in Handan First Hospital were selected. Cancer tissue and adjacent tissue were collected, and patients were divided into a recurrence and metastasis group(n=86) and a non recurrence and metastasis group(n=114) based on postoperative recurrence and metastasis. Immunohistochemistry and real-time fluorescence quantitative polymerase chain reaction were used to collect and analyze the protein expression and relative expression levels of KMT2D, ANXA7, and CMTM6. General clinical data were collected and analyzed. Logistic regression was used to analyse factors influencing postoperative recurrence and metastasis. The receiver operating characteristic curve was plotted to analyze the diagnostic value of KMT2D, ANXA7, and CMTM6 for postoperative recurrence and metastasis in patients. Results The positive expression rates of KMT2D, ANXA7, and CMTM6 proteins in colon cancer tissues were higher than those in adjacent tissues(P<0.05). The proportions of TNM stage Ⅲ+Ⅳ, low differentiation degree, nerve invasion and the relative expression levels of KMT2D, ANXA7, and CMTM6 mRNAin the recurrence and metastasis group were higher than those in the non recurrence and metastasis group(P<0.05). TNM stage Ⅲ+Ⅳ, low differentiation degree, nerve invasion, and elevated mRNA levels of KMT2D, ANXA7, and CMTM6 were all risk factors for postoperative recurrence and metastasis in patients(P<0.05). The area under curve of combination of KMT2D, ANXA7, and CMTM mRNA in predicting postoperative recurrence and metastasis was higher than that of KMT2D(Z=3.233, P=0.001), ANXA7(Z=2.923, P=0.004), and CMTM6(Z=2.037, P=0.042) predicted separately. Conclusion KMT2D, ANXA7, CMTM6 mRNA and protein are upregulated in colon cancer and have a certain relationship with recurrence and metastasis after radical surgery. |
